Overview
Laboratory
Main laboratory of approximately 3000 square feet (HSE 1246) containing 7 dark rooms for microscopy and facilities for rodent surgery/physiology, molecular genetics, organic synthesis (3 fume hoods) and drug discovery. Additional optics rooms located in 11 HSW (120 square feet) and SURGE 215 (120 square feet).
Animal
Animals are housed in the UCSF vivarium under the care of staff veterinarians. The cage fees cover daily animal checks / cage cleaning by technical staff under the supervision of veterinarians.
Computer
The laboratory is equipped with multiple PC and Mac computers interfaced to instrumentation, and for word processing / data analysis, as well as laser/inkjet printers, etc.
Office
Dr. Verkman has a private office with space for small conferences.
Other
An excellent departmental machine shop is available ($ 50 / hr for machining, consultation & design free) for instrument construction, as well as an electronics shop.
Major Equipment
The laboratory is fully equipped for state-of-the art microscopy, rodent surgery/physiology, molecular genetics, high-throughput screening, and organic synthesis. Major equipment items in the laboratory are listed below.
Microscopy / optics instrumentation
- Three Nipkow wheel confocal microscopes, two with rapid z-axial scanning
- Laser sources (two 8 watt Ar lasers, titanium-sapphire femtosecond laser, 5 watt Nd/YAG laser)
- Home-built photobleaching recovery apparatus with microsecond time resolution
- Time-resolved fluorescence spectroscopy/microscopy (SLM 48000 frequency-domain hardware)
- Two home-built fluorescence correlation microscopes (four APDs, EG & G autocorrelators)
- Home-built multi-angle total internal reflection fluorescence microscope
- Eight inverted fluorescence microscopes for imaging/kinetics (5 cooled CCD detectors, multiple PMTs)
- Four Nikon fluorescence stereo zoom microscopes / Nikon DIC brightfield microscope
- Two fluorescence single particle tracking microscopes with TIRF and deep-cooled CCD cameras
High-throughput screening for drug discovery
Biomek integrated workstation, including automated incubator and stations for liquid handling, plate washing, bar code reading, mixing, delidding, and 3 BMG fluorescence platereaders with syringe pumps; ~250,000 drug-like small molecules stored at -20 oC; Waters LC/mass spectrometer
Rodent physiology
Three mouse physiology/surgery stations, with Biopak physiological recorders, stereotaxic frames, monitoring equipment, and surgical stereomicroscopes. Home-built apparatus for measurements of ABRs, ERGs and VEPs. Mouse ventilators and monitoring devices
Electrophysiology
Six Ussing chamber set-ups; Oocyte voltage clamp; Patch-clamp
